The immunochemistry of toxins and toxoids; the solubility of staphylococcal toxin in methanol-water mixtures under controlled conditions of pH, ionic strength, and temperature.

چکیده
The success encountered in the purification of tetanal toxin (1) and toxoid (2, 3), of diphtherial toxoid (4), and of soluble pertussal antigens (5) warranted attempts to purify other toxins and toxoids by the multiphase system of protein fractionation. The system employs the precise adjustment of methanol concentration, pH, ionic strength, and protein concentration under rigidly controlled conditions of temperature in order to separate or isolate proteins. Previous attempts to purify and concentrate staphylococcal toxin have met with varying degrees of success. Holt (6) employed ammonium sulfate to concentrate staphylococcal toxin. Burnet and Freeman (7) precipitated the toxin with acetic acid, while Boivin and Izard (8) used trichloroacetic acid. It is the object of the present communication to report studies on the solubility of staphylococcal toxin in methanol-water mixtures. Subsequent publications will deal with the actual isolation and characterization of the toxin.

متن کاملThe Immunochemistry of Toxins and Toxoids
Methods for the purification and crystallization of tetanal toxin are described. The methods consist of the multiphase fractionation system involving methanol as the organic precipitating agent under controlled conditions of pH, ionic strength, protein concentration, and temperature. Crystalline tetanal toxin has an electrophoretic mobility of 2.8 x 10(-5) in veronal buffer of 0.1 ionic strengt...
متن کاملThermodynamic Study of Water Activity of Single Strong Electrolytes
Today, due to the natural decline of oil exploitation, the use of methods of oil recovery, has made significant progress. However, these methods are accompanied by accumulation and deposition of mineral deposits in oil field installations. In the present study, aqueous solutions, strontium sulfate, barium sulfate, manganese sulfate and nickel sulfate are studied, in terms of EUNIQUAC model and ...
متن کاملThe effect of dielectric constant and ionic strength on the solubility of lithium carbonate at 25.0 °C in thermodynamic view
Using the Solvent Evaporating and Atomic Emission Spectrophotometric methods, the solubilitiesof Li2CO3 were determined in various mixed- solvent "water + ethanol" with various mass percentof ethanol, and in the presence of NaNO3 in various molarities at 25.0°C . The results wereinterpreted in terms of mean ionic activity coefficient and ion association phenomenon.
متن کاملThermodynamic study of ion- association in KNO3 solution in the mixed solvent (water +methanol)
The solubility of KNO3 in the mixed solvent (water 85%+methanol 15%) were determined by using solvent evaporating method at different temperatures (25, 30, 40, 50, 60 ;C). In addition the equilibrium constant of ion pair formation, KIP, for K+NO3 – ion-pair on the basis of Fuoss contact ion pair model was calculated. Upon choosing the extended Debye-Hückel model for estimating the mean activity...
